Real quick…
I have a new EHX OD Glove and an 80’s vintage MXR Dyna Comp.
I have figured out from other posts that the comp should precede the dirt but I wonder if there are any settings I should consider to get a sustained, smooth, wailing tone to augment the presets on my Line 6 Spider III.

Yes the glove should go after the comp, though probably you don’t want to use both at the same time. Distortion/Overdrive pedals already compress the signal. Use it for when you play clean.
As far as the presets on your Spider 3; presets with external pedals is way different than using internal effects. I have a Vox Valvetronix modeling amp and tweak the amp to the pedal, that gives me the best results.
